目的探讨脂联素(ADPN)对高糖环境下肾小球系膜细胞(MCs)增殖及细胞间粘附分子1(ICAM-1)表达的影响。方法将MCs分为三组:①正常组(5.5mmol/L葡萄糖),②高糖组(25mmol/L葡萄糖),③ADPN组(25mmol/L葡萄糖,2.5ug/mlADPN)运用MTT测定(24h,,后MCs的48h96h)增殖情况;再将MCs分为三组:①正常组(5.5mmol/L葡萄糖),②高糖组(25mmol/L葡萄糖),③ADPN组(25mmol/L葡萄糖2.5,,,,,57.5101520ug/mlADPN),作用48小时后运用ELISA法测定各组MCs上清液中ICAM-1的含量。结果高糖组抑制细胞增殖,加入ADPN后促进细胞增殖。ICAM-1在高糖组表达较高,加入低浓度ADPN对于ICAM-1表达影响不大,~20mg/LADPN组和高糖组相比,10ICAM-1的表达下降,差异有统计学差异(P<0.05),且呈剂量依赖性。结论低浓度ADPN可以促进MCs增殖;中等以上浓度ADPN可以抑制ICAM-1在MCs的表达。提示ADPN可能通过阻抑ICAM-1表达发挥对DN的保护作用。
Objective To investigate the effect of adiponectin (ADPN) on the proliferation and the expression of intercellular adhesion molecule-1 (ICAM-1) in glomerular mesangial cells (MCs) under high glucose conditions. Methods MCs were divided into three groups: ① normal group (5.5mmol / L glucose), ② high glucose group (25mmol / L glucose), ③ADPN group (25mmol / L glucose, 2.5ug / mlADPN) Then MCs were divided into three groups: ① normal group (5.5mmol / L glucose), ② high glucose group (25mmol / L glucose), ③ADPN group (25mmol / L glucose 2.5 ,,,, , 57.5101520ug / mlADPN). After 48 hours, the content of ICAM-1 in MCs supernatant of each group was determined by ELISA. Results High glucose group inhibited cell proliferation, and added ADPN to promote cell proliferation. The expression of ICAM-1 in high glucose group was higher than that in low glucose group, while the low concentration of ADPN had little effect on the expression of ICAM-1. Compared with high glucose group, the expression of ICAM-1 in 10mg / LADPN group was significantly decreased (P <0.05) and in a dose-dependent manner. CONCLUSION: Low concentration of ADPN can promote the proliferation of MCs. Middle concentration of ADPN can inhibit the expression of ICAM-1 in MCs. These results suggest that ADPN may exert a protective effect on DN by inhibiting the expression of ICAM-1.
