目的:人脐带间充质干细胞用PKH26标记,探讨人脐带间充质干细胞在大鼠化疗性卵巢早衰模型中的卵巢迁移示踪。方法:提取并培养人脐带间充质干细胞,按PKH26标记程序进行细胞标记,透射电镜观察标记组和未标记组细胞的超微结构,测定细胞增殖、周期、凋亡情况。荧光显微镜下观察PKH26标记的人脐带间充质干细胞移植后的卵巢情况。结果:标记后细胞的形态无明显差别,均为成纤维细胞样,细胞增殖未见明显影响,细胞生长状态良好,PKH26标记阳性的细胞主要存在卵巢血管中。结论:PKH标记技术可用于人脐带间充质干细胞移植治疗化疗所致卵巢早衰中的归巢、分化、增殖的研究。
Objective: Human umbilical cord mesenchymal stem cells were labeled with PKH26 to investigate the migration of human umbilical cord mesenchymal stem cells in ovarian cancer model of premature ovarian failure in rats. Methods: Human umbilical cord mesenchymal stem cells (MSCs) were extracted and cultured. Cell labeling was performed by PKH26 labeling. The ultrastructures of labeled and unlabeled cells were observed by transmission electron microscopy. Cell proliferation, cell cycle and apoptosis were measured. The ovarian tissue after PKH26-labeled human umbilical cord mesenchymal stem cells were observed under a fluorescence microscope. Results: There was no significant difference in the morphology of the cells after labeling. All of the cells were fibroblast-like, the cell proliferation was not significantly affected, and the cell growth was good. PKH26-positive cells mainly existed in the ovarian blood vessels. Conclusion: PKH labeling technique can be used to study the homing, differentiation and proliferation of human umbilical cord mesenchymal stem cells in the treatment of premature ovarian failure induced by chemotherapy.
