目的研究安胰颗粒对重症胰腺炎大鼠NF-κB活化的影响。方法将120只SD大鼠分为正常组、模型(SAP)组、IL-10组和安胰颗粒组,每组又分为3,6,12h三个亚组。模型组予腹腔内注射6%的L-Arginine溶液150mg/100g,1次/1h,共3次,诱导SAP。IL-10组予腹腔注射10000U rhIL-10预处理后诱导SAP。安胰颗粒组连续3天给予安胰颗粒水溶液灌胃后诱导SAP。摘取胰腺组织予以病理学评分并通过免疫组化检测胰腺组织NF-κBp65的表达。结果病理学评分模型组显著高于正常组(P<0.01);安胰颗粒组较模型组显著下降(P<0.05)。胰腺组织NF-κBp65各时点表达模型组显著高于正常组(P<0.01);安胰颗粒组较模型组显著下降(P<0.05,P<0.01),12h时点表达较IL-10组明显下降(P<0.05)。结论安胰颗粒能有效改善重症胰腺炎大鼠胰腺损伤,抑制胰腺组织NF-κBp65的表达可能是其作用机制之一。
Objective To study the effect of AnYi granule on activation of NF-κB in severe pancreatitis in rats. Methods 120 Sprague-Dawley rats were divided into normal group, model group (SAP), IL-10 group and An Pan granule group. Each group was divided into three subgroups: 3,6,12h. The model group was intraperitoneally injected with 6% L-Arginine solution 150mg / 100g, 1 times / 1h, a total of 3 times to induce SAP. IL-10 group was intraperitoneally injected 10000U rhIL-10 pretreatment induced SAP. Anan granule group for 3 consecutive days given pancreatic juice aqueous solution after gavage SAP. Pancreatic tissues were harvested for pathological evaluation and the expression of NF-κBp65 in pancreatic tissues was detected by immunohistochemistry. Results The pathological score of the model group was significantly higher than that of the normal group (P <0.01), and that of the Amniotic Granule group was significantly lower than that of the model group (P <0.05). The expression of NF-κBp65 in pancreatic tissue at each time point was significantly higher than that in the normal group (P <0.01); the level of NF-κB p65 in the model group was significantly lower than that in the normal group (P <0.05, P <0.01) Decreased significantly (P <0.05). Conclusion Amniotic Granules can effectively improve pancreatic injury in rats with severe pancreatitis and inhibit the expression of NF-κBp65 in pancreatic tissues.
