目的探讨大鼠烧伤创面引流淋巴液致炎因子(TNF-α)和抗炎因子(IL-4)水平变化。方法大鼠单侧后腿(随机)浸于70℃热水30s,造成约4%总体表面积(TBSA)深Ⅱ度烫伤,对侧后腿浸于22℃热水30s,实验分为烧伤腿组(A组)和对侧未烧伤腿组(B组);单侧腿烧伤大鼠又称为烧伤组,正常组大鼠未做任何处理,实验分为3个时间点:烧伤后6h(10只)、烧伤后24h(10只)、烧伤后72h(10只)。收集双侧髂总淋巴结输出淋巴管内的淋巴液及动脉血。应用酶联免疫吸附法(ELISA)检测TNF-α、IL-4浓度。结果烧伤后6、24和72h的TNF-α及IL-4水平,A组显著高于B组(P<0.05);血浆TNF-α及IL-4水平,烧伤组与正常组比较差异无显著性。结论烧伤能引起局部引流淋巴液致炎因子与抗炎因子水平升高,从而影响局部免疫。
Objective To investigate the changes of the levels of inflammatory cytokines (TNF-α) and anti-inflammatory cytokines (IL-4) in the draining lymph of burn wounds in rats. Methods The unilateral hind legs of rats were randomly immersed in hot water at 70 ℃ for 30 s, resulting in deep second degree scald of about 4% TBSA. The contralateral hind legs were immersed in hot water at 22 ℃ for 30 s. The experiment was divided into burn leg group (Group A) and contralateral non-burn leg group (group B). Unilateral leg burn rats also known as burn group, the normal group of rats without any treatment, the experiment is divided into three time points: 6h after burn (10 Only), 24h after burn (10), 72h after burn (10). Collect bilateral lymphatic fluid and arterial blood from the common iliac lymph nodes. The concentrations of TNF-α and IL-4 were detected by enzyme-linked immunosorbent assay (ELISA). Results The levels of TNF-α and IL-4 at 6, 24 and 72 h after burn were significantly higher in group A than those in group B (P <0.05). There was no significant difference in plasma levels of TNF-α and IL-4 between burn group and normal group Sex. Conclusion Burn can cause local drainage of lymph inflammatory cytokines and anti-inflammatory cytokines, thus affecting the local immunity.
