目的:应用正交模拟法分析护骨胶囊(HG)各药味对该复方药效的贡献值,并对该方不同组合的复方进行药效模拟。方法:采用正交模拟法,将HG 10个药味按照设计的正交表分为13个组(第13组为阴性对照组),通过血清药理学实验得出相应含药血清,采用CCK-8法检测各组血清对成骨细胞(Ob)增殖的影响。DAS3.0软件处理数据,分析各药味的药效贡献值并对复方药效进行模拟。结果:以Ob增殖为指标,经正交模拟法分析,确定HG各药味在复方中的药效贡献值顺序为:淫羊藿>骨碎补>巴戟天>山药>龟甲>当归>熟地黄>续断>杜仲>制首乌;以Ob增殖为指标,对复方药效进行模拟研究,结果显示27个模拟组方预测药效值明显优于阴性对照组,而且原方最好,4个模拟组方接近于原方。结论:正交模拟法分析HG,确定了HG各药味在复方中的药效贡献值,并发现多个组方的模拟药效与原方接近。
OBJECTIVE: To evaluate the contribution of various medicinal herbs of HG capsule to the efficacy of this compound by orthogonal simulation and to simulate the efficacy of different prescriptions. Methods: The orthogonality method was used to divide the 10 medicinal herbs of HG into 13 groups according to the designed orthogonal table (the 13th group was the negative control group). The corresponding drug serum was obtained through the serum pharmacology experiment. The CCK-8 Method to detect the effect of each serum on the proliferation of osteoblasts (Ob). DAS 3.0 software processes the data, analyzes the potency contributions of each drug and simulates the compound efficacy. Results: Ob proliferated as an index, the orthogonal simulation analysis to determine the contribution of HG each medicine in the compound efficacy of the order: Epimedium> Drynaria> Morinda officinalis> yam> shell> Angelica> Rehmannia > Discontinuation> Eucommia> Radix polygoni multiflori; with Ob proliferation as an index, the pharmacodynamics of the compound was studied by simulation. The results showed that the predictive value of pharmacodynamics of 27 simulated groups was significantly better than that of the negative control group, Simulation group side close to the original party. Conclusion: The orthogonally simulated method is used to analyze HG, and the contribution of HG to each compound is determined. It is found that the simulation efficacy of multiple prescriptions is close to the original one.
