目的 探讨转谷氨酰胺酶 3(transglutaminase3,TGM3)基因在喉癌发生中的作用。方法 在 TGM3基因内部及附近选择微卫星标记进行杂合性丢失 (loss of heterozygosity,L OH)分析 ,于 DNA水平间接检测 72例喉癌患者中该基因的缺失 ;并进一步应用 Northern杂交检测 8例喉癌患者配对肿瘤及癌旁正常组织中 TGM3基因的表达差异。结果 4个短串联重复序列 (shorttandem repeats,STR)位点均存在 L OH,其中 D2 0 S17、D2 0 S6 0 7、D2 0 S99和 D2 0 S841位点的 L OH频率分别为 2 5 .76 %、2 0 .0 0 %、38.10 %和 18.75 % ,至少 1个位点出现 L OH的病例占 6 1.11% ,杂合性丢失与临床分期、淋巴结转移和分化程度无显著相关性 ;Northern杂交结果显示 8例喉癌患者 TGM3基因在正常组织中的表达比肿瘤组织显著增强。结论 TGM3基因在喉癌的发生中具有重要作用 ,具体机理有待进一步研究。
Objective To investigate the role of transglutaminase 3 (TGM3) gene in the pathogenesis of laryngeal carcinoma. Methods The microsatellite markers were used to analyze the loss of heterozygosity (LOH) in and around the TGM3 gene, and the deletion of this gene was detected indirectly in 72 laryngeal cancer patients at the DNA level. Northern blotting was used to detect 8 cases Differential expression of TGM3 gene in paired tumors and adjacent normal tissues of laryngeal cancer patients. Results LOH was found in all four short tandem repeats (STR) sites. The LOH frequencies of D2 0 S17, D2 0 S6 0 7, D2 0 S99 and D2 0 S841 were 25.76 %, 20.0%, 38.10% and 18.75%, respectively. The incidence of LOH in at least one locus accounted for 6 1.11%. The loss of heterozygosity was not significantly correlated with clinical stage, lymph node metastasis and differentiation. Northern blot The results showed that in 8 cases of laryngeal cancer patients with TGM3 gene expression in normal tissue was significantly higher than the tumor tissue. Conclusion TGM3 gene plays an important role in the carcinogenesis of laryngeal carcinoma. The exact mechanism remains to be further studied.
