目的:研究黄连解毒汤不同提取部位及小檗碱、黄芩苷、栀子苷、芦丁对小鼠T、B淋巴细胞增殖的影响。方法:建立体外脾T、B淋巴细胞模型,运用MTT比色法分析比较黄连解毒汤的不同提取部位及其主要化学成分与丝裂原(ConA、LPS)共同作用时对小鼠脾T、B淋巴细胞增殖作用的影响。结果黄连解毒汤全方水提部位(H2O部位)、醇提总部位(EtOH部位)、二氯甲烷萃取部位(CH2Cl2部位)和正丁醇萃取部位(n-Bu部位)对未活化或LPS和ConA诱导的脾T、B淋巴细胞具有不同程度的促进或抑制作用。小檗碱、黄芩苷(10μM)能促进未活化小鼠脾淋巴细胞增殖,而栀子苷、芦丁(50μM)对未活化小鼠脾淋巴细胞的增殖作用不明显。对于脂多糖(LPS)或刀豆蛋白A(ConA)诱导的脾淋巴细胞,小檗碱、黄芩苷、栀子苷、芦丁均能抑制其增殖。结论:黄连解毒汤不同提取部位及其中主要化学成分小檗碱、黄芩苷、栀子苷、芦丁对T、B淋巴细胞均有不同程度的增殖作用。
Objective: To study the effects of different extraction sites of berberine, baicalin, geniposide and rutin on the proliferation of T and B lymphocytes in mice. Methods: Spleen T and B lymphocyte models were established in vitro. MTT colorimetric method was used to analyze the effect of different extraction sites and main chemical components of Huanglian Jiedu Decoction on splenic T, B Effect of lymphocyte proliferation. Results The results showed that the extract of Huanglian Jiedu Decoction (H 2 O) site, the EtOH site, the dichloromethane extract (CH2Cl2) and the n-butanol extract (n-Bu site) Spleen T, B lymphocytes have different degrees of promotion or inhibition. Berberine and baicalin (10μM) could promote the proliferation of splenic lymphocytes in non-activated mice, while geniposide and rutin (50μM) had no significant effect on the proliferation of splenic lymphocytes in non-activated mice. For spleen lymphocytes induced by lipopolysaccharide (LPS) or concanavalin A (ConA), berberine, baicalin, geniposide and rutin could all inhibit their proliferation. CONCLUSION: Berberine, baicalin, geniposide and rutin in different extraction parts of Huanglian Jiedu Decoction and its main chemical components all have different degrees of proliferation on T and B lymphocytes.
