目的观察复方贞术调脂胶囊(FTZ)对代谢综合征(MS)大鼠胰岛素抵抗和胰岛素信号磷脂酰肌醇-3-激酶(PI-3K)的影响,探讨FTZ治疗MS和改善胰岛素抵抗的作用机制。方法采用高热量饲料喂养复制MS大鼠模型,经FTZ治疗8周,测量体重、腹围、血脂、空腹血糖和胰岛素水平,RT-PCR检测脂肪组织PI-3Kp85亚基的mRNA表达水平。结果 FTZ可降低MS大鼠血清TG、TC、空腹血糖、胰岛素及胰岛素抵抗指数,升高血清HDL-C水平、减轻体重和肥胖,上调大鼠脂肪组织PI-3Kp85 mRNA的表达。结论 FTZ可调节MS大鼠糖脂水平,改善胰岛素抵抗,对MS具有防治作用。FTZ上调胰岛素信号PI-3K,可能是其治疗MS和改善胰岛素抵抗的作用机制之一。
Objective To observe the effects of Fufang Zhenzhu Tiaozhi Capsule (FTZ) on insulin resistance and insulin signal phosphatidylinositol 3-kinase (PI-3K) in rats with metabolic syndrome (MS) and explore the effects of FTZ on MS and insulin resistance Mechanism. Methods The MS rat model was fed with high calorie diet. After FTZ treatment for 8 weeks, body weight, abdominal circumference, blood lipids, fasting blood glucose and insulin levels were measured. The mRNA expression of PI-3Kp85 subunit in adipose tissue was detected by RT-PCR. Results FTZ could decrease serum TG, TC, fasting blood glucose, insulin and insulin resistance index, increase serum HDL-C level, reduce body weight and obesity, and up-regulate the expression of PI-3Kp85 mRNA in rat adipose tissue. Conclusion FTZ can regulate the level of glycolipid in MS rats, improve insulin resistance and prevent and treat MS. FTZ upregulates insulin signaling PI-3K, which may be one of the mechanisms of its treatment of MS and improve insulin resistance.
