[目的]探讨舒肝活血化痰方抗非酒精性脂肪性肝炎(NASH)大鼠脂质过氧化的作用机制。[方法]采用高脂饮食复制NASH大鼠模型,用舒肝活血化痰方治疗,以东宝肝泰为对照,观察其对模型大鼠肝功能、血脂、肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)、羟脯氨酸(Hyp)水平及细胞色素P450 2El(CYP2E1)活性的影响。[结果]舒肝活血化痰方显著改善肝功能,降低血清总胆固醇、三酰甘油及低密度脂蛋白胆固醇的同时升高高密度脂蛋白胆固醇水平,并显著降低肝组织Hyp,抑制肝组织CYP 2E1的活性。[结论]舒肝活血化痰方可有效干预大鼠NASH,抑制肝组织CYP 2E1的表达及与其密切相关的脂质过氧化是该方干预作用的主要机制之一。
[Objective] To explore the mechanism of Shugan Huoxue Huatan Decoction on lipid peroxidation in nonalcoholic steatohepatitis (NASH) rats. [Methods] A high-fat diet was used to replicate the NASH rat model and treated with Shugan Huoxue Huatan Decoction. The liver was taken as the control and the liver function, serum lipids, and liver tissue superoxide dismutase (SOD) of the model rats were observed. Activity, malondialdehyde (MDA), hydroxyproline (Hyp) levels, and cytochrome P450 2El (CYP2E1) activity. [Results] Shugan Huoxue Huayu Decoction significantly improved liver function, reduced serum total cholesterol, triacylglycerol, and low-density lipoprotein cholesterol and increased high-density lipoprotein cholesterol levels, significantly reduced liver tissue Hyp, and suppressed liver tissue CYP Activity of 2E1. [Conclusion] Shugan Huoxue Huayu Decoction can effectively intervene NASH in rats, inhibiting the expression of CYP 2E1 in liver tissue and its closely related lipid peroxidation is one of the main mechanisms of this intervention.
