利用标记膜脂及膜蛋白质的荧光标记物DPH和外N-(3芘)马来酰亚胺标记不同浓度细胞刺激物苯并[α]芘、佛波酯及氧自由基作用下的NIH3T3细胞,发现高剂量的刺激物对膜脂损伤显著,低剂量的刺激物虽对细胞膜脂未造成明显损伤,但已引起膜蛋白质构象改变。提示以上刺激物对膜结构的影响可能与影响细胞正常的调控导致细胞的无限生长有关,进而在致、促癌中发挥作用。
NIH3T3 cells were labeled with different concentrations of benzo [a] pyrene, phorbol ester and oxygen free radicals by DPH and N- (3pyrene) maleimide labeled with membrane lipid and membrane protein It was found that the high doses of stimuli had significant damage to membrane lipids, although low doses of stimuli did not cause significant damage to the cell membrane lipids, but caused changes in membrane protein conformation. It is suggested that the above effects of stimulus on the membrane structure may be related to the influence of normal regulation of cells leading to the unlimited growth of cells, thereby contributing to promoting and promoting cancer.
