目的:比较3首治疗黄疸的经典方剂对奈异硫氰酸脂(ANIT)诱发实验性肝内胆汁淤积大鼠肝肾功能变化的影响。方法:一次性给予100 mg.kg-1 ANIT复制大鼠肝内胆汁淤积模型;于造模后次日开始,各药物组分别ig茵陈蒿汤、茵陈五苓散和大柴胡汤(分别含生药9.17,3,16 g.kg-1);模型组和赋形剂组给予等体积生理盐水,连续3 d。至造模72 h时处死取材。结果:与赋形剂组比较,模型组TP,CHO含量无明显变化,Alb含量,LDH活性显著降低;ALT,AST,ALP,GGT活性,Glo,TBil,TBA,Cre含量显著增高(P<0.01,P<0.05);Alb含量明显降低(P<0.01);与模型组比较,茵陈蒿汤、茵陈五苓散和大柴胡汤均可显著增加Alb,TP含量(P<0.01),显著降低ALT,AST,ALP,GGT活性,TBil,TBA,Cre含量(P<0.01),茵陈蒿汤在降低TBil方面显著优于茵陈五苓散(P<0.05)。结论:茵陈蒿汤、茵陈五苓散和大柴胡汤均能有效改善ANIT所诱发肝内胆汁淤积大鼠模型肝肾功能的改变。
Objective: To compare the effects of three classic prescriptions of jaundice on the changes of liver and kidney function in rats with intrahepatic cholestasis induced by Nano-isothiocyanate (ANIT). Methods: Intrahepatic cholestasis model was established by intraperitoneal injection of 100 mg.kg-1 ANIT once a day. The rats in each group were treated with Yinchenhao Decoction, Yinchen Wuling Powder and Dachaihu Decoction 9.17,3,16 g.kg-1); model group and vehicle group were given equal volume of normal saline for 3 days. The model was sacrificed at 72 h. Results: Compared with the vehicle group, the levels of TP and CHO did not change significantly in the model group, while the contents of Alb and LDH decreased significantly. The activities of ALT, AST, ALP and GGT, Glo, TBil, TBA and Cre significantly increased (P <0.01, P <0.05). The content of Alb was significantly decreased (P <0.01). Compared with the model group, the contents of Alb and TP of Yinchenhao decoction, Yinchen Wuling powder and Dachaihu decoction increased significantly , ALP, GGT activity, TBil, TBA and Cre (P <0.01). Yinchenhao Decoction was significantly better than Yinchen Wuling Powder in reducing TBil (P <0.05). Conclusion: Yinchenhao decoction, Yinchen Wuling powder and Dachaihu decoction all can effectively improve the changes of liver and kidney function in rats with intrahepatic cholestasis induced by ANIT.
