目的:利用免疫组化法研究温胆汤、化瘀温胆汤对糖耐量低减(IGT)大鼠脂肪中PI3K、PKB、GSK-3β蛋白表达的影响。方法:40只SD大鼠随机分为空白对照组、IGT模型组、温胆汤组和化瘀温胆汤组,喂养24周后,取脂肪采用免疫组化法测定PI3K、PKB、GSK-3β蛋白含量。结果:与空白对照组比较,IGT组PI3K、PKB蛋白含量显著降低(P<0.05),GSK-3β蛋白含量显著升高(P<0.05)。与IGT组比较,温胆汤、化瘀温胆汤均可显著升高PI3K、PKB蛋白含量(P<0.05),降低GSK-3β蛋白含量(P<0.05)。结论:温胆汤、化瘀温胆汤可以不同程度的作用于IGT大鼠脂肪的PI3K通路。
OBJECTIVE: To study the effect of Wendan Decoction and Huayu Wendan Decoction on the expression of PI3K, PKB and GSK-3β in fat of rats with impotence (IGT) by immunohistochemistry. Methods: Forty SD rats were randomly divided into blank control group, IGT model group, Wendan decoction group and Huayu Wendan decoction group for 24 weeks. The levels of PI3K, PKB and GSK-3β were determined by immunohistochemistry Protein content. Results: Compared with the blank control group, the protein levels of PI3K and PKB in IGT group were significantly decreased (P <0.05) and GSK-3β protein content was significantly increased (P <0.05). Compared with IGT group, Wen Dan Tang and Huayu Wendan Decoction could significantly increase the contents of PI3K and PKB protein (P <0.05) and reduce the content of GSK-3β protein (P <0.05). Conclusion: Wendan Decoction and Huayu Wendan Decoction can affect the PI3K pathway of adipose tissue in IGT rats to varying degrees.
