目的探讨全反式维甲酸对大鼠C6脑胶质瘤细胞增殖抑制作用及细胞周期素Cy- clin E和凋亡相关基因Caspase-3 mRNA表达的影响。方法噻唑蓝(MTT)比色法检测全反式维甲酸(ATRA)在不同浓度(1、2、4、8、16μmol/L)、不同作用时间(24、48、72 h)处理大鼠胶质瘤C6细胞系后,对细胞增殖抑制率的变化;逆转录-聚合酶链反应(RT-PCR)法检测细胞周期素Cyclin E和Caspase-3 mRNA在24、48、72 h不同时间点表达变化。结果ATRA对C6细胞有增殖抑制作用,具有时间和浓度依赖性;药物作用组与无药物作用组之间及药物作用各组之间的OD值和抑制率比较差异有统计学意义(P<0.01)。随ATRA作用时间的延长,细胞周期素Cyclin E mRNA表达逐渐下降,与细胞凋亡相关基因Caspase-3 mRNA表达逐渐增高(P<0.01)。结论ATRA抑制C6脑胶质瘤细胞发生增殖,其机制可能与细胞周期素Cyclin E表达下调,凋亡相关基因Caspase-3上调有关。
Objective To investigate the effects of all-trans retinoic acid on the proliferation of rat C6 glioma cells and the expression of Cy-clin E and the apoptosis related gene Caspase-3 mRNA. Methods MTT assay was used to detect the expression of all-trans retinoic acid (ATRA) at different concentrations (1, 2, 4, 8 and 16μmol / L) The expression of cyclin E and Caspase-3 mRNA was detected by RT-PCR at 24, 48, and 72 h after transfection Variety. Results ATRA inhibited proliferation of C6 cells in a time and concentration-dependent manner. There was significant difference in OD value and inhibition rate between drug-treated group and non-drug-treated group and drug-treated group (P <0 .01). With the prolongation of ATRA effect, the expression of cyclin E mRNA gradually decreased and the expression of Caspase-3 mRNA increased (P <0.01). Conclusions ATRA can inhibit the proliferation of C6 glioma cells, which may be related to the down-regulation of cyclin E expression and up-regulation of Caspase-3.
