目的:观察地骨皮醇提取液对2型糖尿病大鼠肾脏COX-2表达的影响,探讨其对糖尿病肾病大鼠肾脏保护作用的机制。方法:SD大鼠125只,随机抽取10只为正常对照组。以STZ加高脂饲料饲养3周复制2型糖尿病肥胖大鼠模型,随机分为模型组、罗格列酮组、地骨皮醇提低剂量组、地骨皮醇提高剂量组,每组10只。模型组和正常组给予生理盐水,各实验组分别给予相应药物灌胃,治疗7周后免疫组化法检测肾脏COX-2表达。结果:罗格列酮组和地骨皮醇提组COX-2表达与模型组比较明显改善,差异均有统计学意义(P<0.01),但与正常对照组之间仍有明显差异(P<0.01)。地骨皮醇提高剂量组与罗格列酮组比较差异无统计学意义(P>0.05)。结论:地骨皮醇提取液可以降低2型糖尿病肥胖大鼠肾脏COX-2的表达,其中高剂量组优于低剂量组,与罗格列酮组疗效相当。
OBJECTIVE: To observe the effect of Digupi alcohol extract on the expression of COX-2 in the kidney of type 2 diabetic rats and to explore its protective mechanism on diabetic nephropathy rats. Methods: 125 Sprague - Dawley rats were randomly divided into normal control group and control group. The model rats were fed with STZ plus high-fat diet for 3 weeks and were randomly divided into model group, rosiglitazone group, digoxin-lowering dose group and digroup-alcohol-raising dose group only. The model group and the normal group were given normal saline, and the experimental groups were given the corresponding drug gavage. After 7 weeks of treatment, the expression of COX-2 in the kidney was detected by immunohistochemistry. Results: The expression of COX-2 in Rosiglitazone and Digupill groups significantly improved compared with the model group (P <0.01), but there was still significant difference between the rosiglitazone group and the control group (P <0.01). There was no significant difference in the dose of digoxin from the rosiglitazone group (P> 0.05). CONCLUSION: Digoxigenin can decrease the expression of COX-2 in kidney of type 2 diabetic obese rats. The high-dose group is superior to low-dose group and has the same effect with rosiglitazone group.
