目的建立测定生知母和盐知母中芒果苷和菝葜皂苷元的方法。方法分别采用HPLC-UV法和HPLC-ELSD法测定芒果苷和菝葜皂苷元。测定芒果苷的色谱条件:Kromasil柱(250mm×4.6mm,5μm);水-乙腈-0.85%磷酸(100∶25∶5)为流动相;体积流量为0.6mL/min;柱温为30℃;检测波长为258nm。测定菝葜皂苷元的色谱条件:Kromasil柱(250mm×4.6mm,5μm);甲醇-水(95∶5)为流动相;体积流量为1.0mL/min;柱温30℃;用蒸发光散射检测器检测,漂移管温度85℃。结果盐知母中芒果苷和菝葜皂苷元的量均有所升高。结论本方法可用于生知母和盐知母中芒果苷和菝葜皂苷元的测定,并为其全面质量控制提供参考。
Objective To establish a method for the determination of mangiferin and smilagenin in Zhimu and Yanzhimu. Methods Mangiferin and Smilagenin were determined by HPLC-UV and HPLC-ELSD, respectively. The chromatographic conditions were as follows: Kromasil column (250 mm × 4.6 mm, 5 μm); water-acetonitrile-0.85% phosphoric acid (100:25:5) as mobile phase; volume flow rate of 0.6 mL / The detection wavelength is 258nm. The chromatographic conditions were: Kromasil column (250mm × 4.6mm, 5μm); methanol-water (95: 5) as mobile phase; volume flow rate of 1.0mL / min; Device detection, drift tube temperature 85 ℃. Results The contents of mangiferin and sarsasapogenin in Zhimu were all increased. Conclusion The method can be applied to the determination of mangiferin and sarsasapogenin in Zhimu and Yanzhimu, and provide a reference for its total quality control.
