目的 探索用双重腺病毒载体介导bax基因对大肠癌进行基因治疗。方法 双重腺病毒载体介导bax表达 ,相差显微镜、MTT比色法、流式细胞仪观察bax基因对HT 2 9的作用 ,WesternBlot检测Bax蛋白的表达。结果 Ad/GT bax能有效地引起HT 2 9形态学的改变 ,当加入Ad/PGK GV16后 ,该作用更为显著 ,呈典型的凋亡形态学特征 ,其生长抑制率及凋亡诱导率分别为 5 7.4%和 2 8.2 % ,与Ad/GT bax组的 2 9.2 %和 12 .6 %相比 ,差异有显著性 (P <0 .0 5 )。Ad/GT bax与Ad/PGK GV16同时应用能引起Bax蛋白大量表达 ,而单用Ad/GT bax ,则Bax蛋白表达量较低。结论 bax基因具有有效诱导人大肠癌细胞株HT 2 9凋亡的作用
Objective To explore the gene therapy of colorectal cancer with bax gene mediated by dual adenovirus vector. Methods Double adenovirus vector was used to mediate bax expression. Phase contrast microscopy, MTT colorimetric assay and flow cytometry were used to observe the effect of bax gene on HT 2 9. Western blot was used to detect the expression of Bax protein. Results Ad / GT bax could effectively induce the morphological changes of HT 2 9. This effect was more pronounced when Ad / PGK GV16 was added, showing typical morphology of apoptotic cells. The growth inhibitory rate and apoptosis induction rate were respectively Compared with 5.22% and 5.22% of the Ad / GT bax group, the difference was significant (P <0.05). The simultaneous application of Ad / GT bax and Ad / PGK GV16 caused a large amount of Bax protein expression, while Ad / GT bax, Bax protein expression was low. Conclusion bax gene can effectively induce the apoptosis of human colorectal cancer cell line HT 2 9
