本研究工作首次应用微核测定技术快速评价胱胺和H_2O_2在照射前分别或合并使用时,它们对带瘤小鼠的骨髓细胞和艾氏腹水癌细胞染色体辐射损伤的影响。微核测定的结果表明: (1)照前口服或腹注胱胺对受一次或分次照射的腹水癌小鼠骨髓细胞的染色体损伤均有显著性的保护作用,而对其癌细胞的染色体损伤无明显的防护效果。(2)H_2O_2能够提高腹水癌细胞染色体辐射损伤的敏感性,而对其骨髓细胞的染色体损伤无明显的影响。(3)胱胺和H_2O_2合并使用时,H_2O_2不降低胱胺对骨髓细胞的防护作用,而胱胺却能削弱H_2O_2对癌细胞的致敏作用。
This study was the first to rapidly evaluate the effect of cystamine and H 2 O 2 on the chromosome damage of bone marrow cells and Ehrlich ascites carcinoma cells before and after irradiation with micronucleus assay. The results of micronucleus assays showed that: (1) Oral or intraperitoneal injection of cystamine before irradiation significantly protects the chromosomes of bone marrow cells of ascites carcinoma mice with single or fractional exposure to chromosome damage. Damage has no obvious protective effect. (2) H_2O_2 can increase the sensitivity of chromosome damage in ascites cancer cells, but has no obvious effect on the chromosome damage of bone marrow cells. (3) When cystamine and H 2 O 2 were combined, H 2 O 2 did not reduce the protective effect of cystamine on bone marrow cells, while cystamine could attenuate the sensitization of H 2 O 2 to cancer cells.
