目的:研究天晴甘平对化疗药物致肝损伤大鼠肝功能的保护作用。方法:选择C57BL/6大鼠作为研究对象,适应性饲养后随机分为对照组、顺铂组以及干预组,顺铂组和干预组接受顺铂腹腔注射,干预组接受天晴甘平灌胃。顺铂给药后,检测3组大鼠血清中氨基转移酶的含量以及肝脏组织中能量代谢相关酶、炎症介质、凋亡基因的含量。结果:给药后1d、2d、3d、4d,顺铂组大鼠血清中ALT、AST的含量均高于对照组,干预组大鼠血清中ALT、AST的含量均低于对照组,均P<0.05;处死大鼠收集肝脏组织,顺铂组大鼠肝脏组织中SDH、Na+-K+-ATP酶的mRNA含量低于对照组,TNF-α、IL-1β、IL-6、CXCL8的蛋白含量以及caspase-3、caspase-8、caspase-9的mRNA含量高于对照组,均P<0.05;干预组大鼠肝脏组织中SDH、Na+-K+-ATP酶的mRNA含量高于顺铂组,TNF-α、IL-1β、IL-6、CXCL8的蛋白含量以及caspase-3、caspase-8、caspase-9的mRNA含量均低于顺铂组,均P<0.05。结论:天晴甘平对顺铂所致肝损伤大鼠的肝功能具有保护作用,具体表现为改善细胞能量代谢、减轻炎症反应、抑制凋亡过程。
Objective: To study the protective effects of Ganqing Ganping on liver function in rats induced by chemotherapy drugs. Methods: C57BL / 6 rats were selected as experimental subjects and randomly divided into control group, cisplatin group and intervention group. Cisplatin group and intervention group received cisplatin intraperitoneal injection. . After cisplatin administration, the content of aminotransferase and the enzyme related to energy metabolism, inflammatory mediators and apoptotic genes in liver tissues were detected. Results: The levels of ALT and AST in the serum of cisplatin group were significantly higher than those of the control group on the 1st, 2nd, 3rd, 4th day after the administration, the contents of ALT and AST in the intervention group were lower than those in the control group <0.05; The liver tissue of the rats was sacrificed, and the content of SDH, Na + -K + -ATPase in the liver tissue of the cisplatin group was lower than that of the control group. The protein contents of TNF-α, IL-1β, IL-6 and CXCL8 (P <0.05). The content of mRNA of SDH and Na + -K + -ATPase in hepatic tissue of intervention group was higher than that of cisplatin group and TNF-α , The contents of IL-1β, IL-6 and CXCL8, as well as the mRNA contents of caspase-3, caspase-8 and caspase-9 were all lower than that of cisplatin group (P <0.05). CONCLUSION: Tiangan Ganping has a protective effect on the liver function of rats with liver injury induced by cisplatin, specifically improving the energy metabolism of cells, reducing the inflammatory reaction and inhibiting the apoptosis process.
