目的研究慢性脑缺血Orexin受体-2(OX2R)的表达。方法通过结扎双侧颈总动脉建立脑缺血模型;用水迷宫进行行为学评价;双标免疫荧光观察OX2R的表达。结果缺血15d时大鼠的学习记忆能力明显减退,1个月、2个月较缺血15d模型组学习记忆能力有所好转。同时,缺血15d时OX2R在神经元的表达明显增高,1个月时OX2R的表达较15d时明显下降,2个月时OX2R的表达再次增高;此外,OX2R在神经纤维亦有表达其变化趋势同神经元。组织学示15d时部分细胞萎缩,1个月时大部分细胞变形萎缩,2月时部分细胞形态恢复正常。结论提示脑缺血的早期OX2R可能有诱导细胞凋亡的作用,而缺血的后期有神经保护作用。
Objective To investigate the expression of Orexin Receptor-2 (OX2R) in chronic cerebral ischemia. Methods The model of cerebral ischemia was established by ligating bilateral common carotid arteries. The behavioral evaluation was performed by water maze. The expression of OX2R was observed by double-labeled immunofluorescence. Results The learning and memory abilities of rats decreased significantly on the 15th day after ischemia, and the learning and memory abilities of the model group at 1 month and 2 months were better than those of the 15th day. At the same time, the expression of OX2R in neurons increased significantly at the 15th day after ischemia, the expression of OX2R at 1 month decreased significantly compared with the 15th day, and the expression of OX2R increased again at 2 months. In addition, OX2R also expressed in nerve fibers With the neurons. Some of the cells showed atrophy on the 15th day of histology, most of the cells degenerated and atrophied at 1 month, and part of the cell morphology returned to normal in February. The conclusion suggests that early OX2R in cerebral ischemia may induce apoptosis, while neuroprotective effect is observed in the later phase of ischemia.
