目的探究海带多糖(Laminaria japonica polysaccharides,LJP)对慢性局部电离辐射致雄性大鼠睾丸细胞损伤的影响。方法将适应饲养7 d的56只Wistar雄性大鼠随机分为正常对照1组、辐射模型3组和LJP干预3组,每组8只;LJP干预3个组给予LJP的生理盐水溶液,其余各组以等量生理盐水灌胃;经灌胃处理7d后,除对照组外的其它6组,以60Coγ射线进行局部照射,除周末两天不照射外,每天1次,每周连续5次,共20次,总剂量每只2.3 Gy;分别在停止照射后1、7和14 d处死大鼠,分离睾丸;采用免疫组化、单细胞凝胶电泳、流式细胞术的方法研究海带多糖对电离辐射致大鼠睾丸组织caspase3蛋白表达量和睾丸细胞DNA损伤及其含量的影响。结果停止辐射1d、7d、14d后LJP各组与对应模型组相比,睾丸组织形态好于对应模型组,睾丸组织caspase3蛋白表达量降低,其彗星拖尾率、尾长减少;HC峰内细胞占睾丸细胞百分比明显低于对应模型组,且LJP 14d组1C、2C、4C峰内细胞占睾丸细胞百分比均显著高于模型14d组。结论 LJP可明显降低辐射所致大鼠睾丸组织caspase 3蛋白的异常表达和各睾丸细胞的DNA损伤,促进损伤细胞的修复。
Objective To investigate the effects of Laminaria japonica polysaccharides (LJP) on testicular cell injury induced by chronic local ionizing radiation in male rats. Methods Fifty-six male Wistar rats, which were adapted to feeding for 7 days, were randomly divided into normal control group (1), irradiation model group (3) and LJP intervention group (n = 8). LJP intervention group was given LJP saline solution, Groups were treated with equal volume of normal saline; After 7 days of intragastric administration, other 6 groups except the control group were irradiated with 60Coγ-ray, except for the two days of the weekend, once a day for five times a week, A total of 20 times, the total dose of 2.3 Gy each; rats were sacrificed at 1, 7 and 14 days after irradiation was stopped, testicular separation; immunohistochemistry, single cell gel electrophoresis, flow cytometry method of kelp polysaccharide Effect of ionizing radiation on the expression of caspase 3 protein and DNA damage and the content of testicular cells in testis of rats. Results Compared with the corresponding model group, the morphological changes of testis tissue in the LJP groups after stopping radiation for 1, 7 and 14 days were lower than those in the corresponding model group. The expression of caspase 3 protein in the testis tissue was decreased and the tailing and tail length of the comets decreased. Accounting for the percentage of testicular cells was significantly lower than the corresponding model group, and LJP 14d group 1C, 2C, 4C peak percentage of testicular cells were significantly higher than the model 14d group. Conclusion LJP can significantly reduce the radiation-induced abnormal expression of caspase 3 protein in rat testis and DNA damage in testicular cells and promote the repair of injured cells.
