目的采用UPLC法测定蒙药清热八味丸中胆酸的含量。方法采用Waters Acquity UPLC BEH C18色谱柱(100 mm×2.0 mm,1.7μm),流动相为甲醇-水-冰醋酸(80∶20∶0.01),流速为0.4 m L·min-1,柱温为30℃;ELSD条件的雾化器温度40℃,漂移管温度85℃,载气流量2.5 L·min-1。结果胆酸的线性范围为0.9016~4.5080μg(r=0.9993),平均回收率为98.9%,RSD=1.9%(n=9)。结论所用方法简便、快速、准确、重复性好,可用于清热八味丸中胆酸的定量分析。
Objective To determine the content of bile acid in Mongolian medicine Bawei pill by UPLC method. Methods Waters Acquity UPLC BEH C18 column (100 mm × 2.0 mm, 1.7 μm) was used. The mobile phase was methanol-water-glacial acetic acid (80:20:0.01) at a flow rate of 0.4 m L · min-1. The column temperature was 30 ℃. The nebulizer temperature of ELSD was 40 ℃, the temperature of drift tube was 85 ℃ and the carrier gas flow was 2.5 L · min-1. Results The linear range of cholic acid was 0.9016 ~ 4.5080μg (r = 0.9993). The average recovery was 98.9% with RSD = 1.9% (n = 9). Conclusion The method is simple, rapid, accurate and reproducible. It can be used for the quantitative analysis of bile acids in Qingwei Bawei pill.
