目的:建立高效液相色谱法测定体外小鼠巨噬细胞摄取吡喹酮含量的方法,为药物新制剂长循环纳米粒的处方筛选提供方法。方法:采用Kromasil 100-5 C_(18)柱(250 mm×4.6mm,5 m),流动相为甲醇水(67:33,v/v);检测波长为264 nm;柱温为30℃;流速为1 mLmin~(-1);外标法定量。结果:吡喹酮在2.0~40.0 gmL~(-1)与峰面积有良好的线性关系,r~2=0.9992,方法回收率:95.4%~109.1%,RSD<10.6%(n=15)。结论:该方法可简便准确地测定体外巨噬细胞摄取吡喹酮的含量。
OBJECTIVE: To establish a method for the determination of praziquantel uptake by macrophages in vitro by high performance liquid chromatography (HPLC), and to provide a method for the prescription screening of long-circulating drug-loaded nanoparticles. Methods: Kromasil 100-5 C 18 column (250 mm × 4.6 mm, 5 m) was used. The mobile phase was methanol water (67:33, v / v). The detection wavelength was 264 nm. The column temperature was 30 ℃. Flow rate of 1 mLmin ~ (-1); external standard method. Results: There was a good linear relationship between peak area and area of praziquantel at 2.0 ~ 40.0 g mL ~ (-1), r ~ 2 = 0.9992. The recovery rate was 95.4% -109.1% with RSD <10.6% (n = 15). Conclusion: This method can be easily and accurately measured in vitro macrophage uptake praziquantel content.
