目的探讨感染性休克时循环内皮细胞(CEC)、血管性血友病因子(vWF)与炎症相关因子的变化及其相关性。方法将SD大鼠32只随机分为空白对照组(n=8只)与感染性休克组(n=24只)。空白对照组注射生理盐水,感染性休克组注射内毒素(LPS,10mg/kg)诱导休克。对照组即刻采血,休克组制模后1h、2h、4h采血(n=8只)。用流式细胞仪检测血液中CEC数量,酶联免疫吸附法(ELISA)检测血液中vWF、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)的含量。结果①与对照组相比,感染性休克组CEC数量、vWF水平明显增加,血清TNF-α、IL-1β水平亦明显升高,差异均有显著性(P<0.01)。②随着休克时间延长,CEC数量及vWF、TNF-α、IL-1β水平持续增加,且有显著性差异(P<0.01)。③CEC数量与TNF-α、IL-1β水平呈显著正相关(r1=0.998,P1=0.002;r2=0.974,P2=0.026),vWF含量与TNF-α、IL-1β水平亦呈显著正相关。结论感染性休克时,血管内皮受损,炎症相关因子可能起关键作用,CECs、vWF的变化与炎症引起的血管内皮损伤有关。i c
Objective To investigate the changes and correlations of circulating endothelial cells (CECs), von Willebrand factor (vWF) and inflammation related factors in septic shock. Methods 32 SD rats were randomly divided into blank control group (n = 8) and septic shock group (n = 24). The blank control group was injected with saline, and the septic shock group was induced with shock by injecting endotoxin (LPS, 10mg / kg). Blood samples were collected immediately in the control group and blood was collected at 1h, 2h and 4h after shock in the shock group (n = 8). The amount of CEC in the blood was detected by flow cytometry. The contents of vWF, TNF-α and IL-1β in the blood were detected by enzyme linked immunosorbent assay (ELISA). Results ① Compared with the control group, the levels of CEC and the level of vWF in septic shock group were significantly increased, and the levels of serum TNF-α and IL-1β were also significantly increased (P <0.01). ② With the prolongation of shock time, the levels of CEC and the levels of vWF, TNF-α and IL-1β continued to increase, with significant difference (P <0.01). There was a significant positive correlation between the number of CEC and the levels of TNF-α and IL-1β (r1 = 0.998, P1 = 0.002; r2 = 0.974, P2 = 0.026). Conclusions In septic shock, endothelial dysfunction and inflammation related factors may play a key role. The changes of CECs and vWF are related to vascular endothelial injury induced by inflammation. i c
