目的:研究柿叶多糖对链脲佐菌素糖尿病小鼠的降血糖作用及其作用机制。方法:将实验小鼠随机分成5组:正常对照组、模型组、格列本脲组(25 mg.kg-1.d-1),柿叶多糖低、高剂量组(150,600 mg.kg-1.d-1)。ig给于药物15 d,并在相应时间采血测定空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素敏感指数(ISI)、丙二醛(MDA)、超氧化歧化酶(SOD)等指标。结果:高剂量的柿叶多糖能较明显的改善糖尿病小鼠的葡萄糖耐量(P<0.01),低、高剂量组均能显著降低糖尿病小鼠的血糖水平(P<0.05),有效降低肝组织MDA值,相应增高SOD活力,并能显著增加胰岛素敏感指数(ISI),改善胰岛素抵抗状态。结论:柿叶多糖具有较强的降血糖作用,可能与其能提高小鼠肝脏抗氧化酶活性、改善胰岛素抵抗等有关。
Objective: To study the hypoglycemic effect and its mechanism of persimmon leaf polysaccharide on streptozotocin-induced diabetic mice. Methods: The experimental mice were randomly divided into 5 groups: normal control group, model group, glibenclamide group (25 mg.kg-1.d-1), persimmon leaf polysaccharide low and high dose group (150 and 600 mg.kg- 1.d-1). ig was given to the drug for 15 days and the blood samples were taken at the corresponding time to determine the fasting blood glucose (FBG), fasting insulin (FINS), insulin sensitivity index (ISI), malondialdehyde (MDA) and superoxide dismutase (SOD) Results: Polysaccharide of persimmon leaf could significantly improve the glucose tolerance of diabetic mice (P <0.01). Both of low and high dose groups could significantly decrease the blood glucose level of diabetic mice (P <0.05) and reduce the hepatic tissue MDA, correspondingly increased SOD activity, and can significantly increase the insulin sensitivity index (ISI), improve insulin resistance status. Conclusion: Persimmon leaf polysaccharide has a strong hypoglycemic effect, which may be related to its ability to increase liver anti-oxidative enzyme activity and improve insulin resistance.
