目的 :探讨原发性肺癌中 bcl- 2、bax和 p5 3m RNA表达水平及其与临床病理特征的关系。方法 :用免疫组织化学 Dako Envision TM二步法、反转录聚合酶链反应 (RT- PCR)方法分别检测5 2例肺癌组织中 bcl- 2、bax、p5 3m RNA表达情况。结果 :5 2例肺癌组织中 bcl- 2、bax蛋白表达阳性率分别为 4 2 .3%、5 9.6 1%。肺癌中 bcl- 2蛋白过表达在低分化癌、 期肺癌中高于高分化癌以及 期肺癌差异有显著性意义 (P<0 .0 1)。 p5 3m RNA、bax蛋白的阳性表达与 bcl- 2蛋白呈负相关。结论 :bcl- 2蛋白在晚期肺癌中过表达 ,p5 3m RNA、bax蛋白在早期肺癌中起着重要作用 ,它们共同参与细胞内死亡和存活信息的平衡状态的调节。
Objective: To investigate the expression of bcl-2, bax and p5 3m RNA in primary lung cancer and its relationship with clinicopathological features. Methods: The expressions of bcl-2, bax and p53mRNA in 52 lung cancer tissues were detected by immunohistochemical Dako Envision TM and reverse transcription-polymerase chain reaction (RT-PCR). Results: The positive rates of bcl-2 and bax in 52 cases of lung cancer were 42.3% and 59.6% respectively. The overexpression of bcl-2 protein in lung cancer was significantly higher than that in well-differentiated cancer and stage lung cancer in poorly differentiated and advanced lung cancer (P <0.01). The positive expression of p5 3m RNA and bax protein was negatively correlated with bcl-2 protein. CONCLUSION: The bcl-2 protein is overexpressed in advanced lung cancer. The p5 3 mRNA and bax protein play an important role in early stage lung cancer. They both participate in the regulation of the balance of intracellular death and survival information.
