黄精多糖对小鼠腹腔源性巨噬泡沫细胞形成的抑制作用

目的:研究黄精多糖对小鼠腹腔源性巨噬泡沫细胞(MFC)形成的抑制作用。方法:体外传代培养细胞,以20 mg/L氧化型低密度脂蛋白(ox-LDL)培养细胞以复制MFC模型。试验分为空白对照(常规培养基)组、模型(常规培养基)组与黄精多糖高、中、低浓度(质量浓度分别为80、40、20 mg/L)组,复制模型的同时给药培养,连续72 h。以油红O染色,显微镜下观察细胞形态,计数10个视野内的细胞总数与MFC数并计算MFC形成率;测定细胞中总胆固醇(TC)、游离胆固醇(FC)含量并计算胆固醇酯(EC)量;Western blot法测定CD36、三磷酸腺苷结合转运子(ABCA)1、胆固醇酰基转移酶(ACAT)1蛋白的表达。结果:空白对照组细胞体积未见明显增大,油红O染色阳性细胞数量少。与空白对照组比较,模型组细胞体积增大,镜下可见细胞红油O染色阳性细胞数量增多,MFC形成率升高,TC、FC、EC含量增加,CD36、ACAT1蛋白表达增强,ABCA1蛋白表达减弱,差异有统计学意义(P<0.01)。与模型组比较,黄精多糖高、中、低浓度组细胞接近梭形,油红O染色阳性细胞数量减少,MFC形成率降低,TC、FC、EC含量减少,CD36、ACAT1蛋白表达减弱,ABCA1蛋白表达增强,差异有统计学意义(P<0.01或P<0.05)。结论:黄精多糖可抑制因ox-LDL诱导的MFC的形成。其机制可能与减少巨噬细胞内胆固醇的含量,下调CD36、ACAT1蛋白,并上调ABCA1蛋白表达有关。 Objective: To study the inhibitory effect of Polygonatum polysaccharides on the formation of mouse peritoneal macrophage foam cells (MFC). Compared with the model group, the cells in high, medium and low concentration groups of Polygonatum sibiricus were close to fusiform, the number of oily red O staining positive cells decreased, the formation rate of MFC reduced, the contents of TC, FC and EC decreased, the expression of CD36 and ACAT1 decreased, The expression increased, the difference was statistically significant (P <0.01 or P <0.05). Conclusion: Polygonatum polysaccharide can inhibit the formation of MFC induced by ox-LDL. The mechanism may be related to reducing the content of cholesterol in macrophages, down-regulating CD36, ACAT1 protein and up-regulating ABCA1 protein expression.
