用正常成年雄性Wistar大鼠46只,分为正常对照组,实验性脾虚组,自然恢复组和四君子汤治疗组.取十二指肠,Bouin液固定,制石蜡切片,进行了以下染色.(1)H.E染色(2)按免疫组织化学PAP法显示粘膜的胃泌素(G)细胞、生长抑素(D)细胞和5-HT细胞.在光镜下按照半定量法将细胞分为强阳性、中等阳性和弱阳性.观察并记录每组动物切片中各级活性细胞数和计算各级细胞的百分比.(3)从正常对照组,脾虚组各随机选择5例动物,对D细胞和5-HT细胞进行显微分光光度计的定量测定.结果表明(1)脾虚组粘膜未见明显的组织学变化.(2)内分泌细胞:与对照组相比,脾虚组G细胞、5-HT细胞内反应物含量减少,表明分泌功能增强.D细胞内反应物含量增多,表明分泌功能减弱.统计学分析表明以上结果有显著性差异(P值<0.01).与自然恢复比较,治疗组接近对照组.本文结果提示,脾虚证的发病与消化道内分泌细胞变化有密切关系;四君子汤对脾虚证有较好的治疗作用.
46 normal adult male Wistar rats were divided into normal control group, experimental spleen deficiency group, natural recovery group and Sijunzi Decoction treatment group. The duodenum was taken and fixed with Bouin solution to make paraffin section. The following staining was performed. 1) HE staining (2) Gastric mucosal gastrin (G) cells, somatostatin (D) cells, and 5-HT cells were visualized by immunohistochemical PAP. The cells were subdivided into semi-quantitative methods under light microscopy. Positive, medium positive, and weak positive. Observe and record the number of active cells at each level in each group of animals and calculate the percentage of cells at all levels. (3) Choose 5 animals randomly from normal control group and spleen deficiency group for D cells and 5-HT cells were quantitatively measured by microspectrophotometry. The results showed that (1) No obvious histological changes were observed in the mucosa of the spleen deficiency group. (2) Endocrine cells: Compared with the control group, the spleen deficiency group G cells, 5-HT The decrease in the amount of intracellular reactants indicates that the secretion function is enhanced. The content of reactive substances in the D cell increases, indicating that the secretion function is weakened. Statistical analysis shows that the above results have significant differences (P value < 0.01). Compared with natural recovery, the treatment group is close The control group.
