用高效液相色谱法(HPLC)测定5名多动综合征患儿口服匹莫林片剂后的体浓度。约服药后2h,其血药浓度和唾药浓度均可到达峰值。血药和唾药的消除半减(衰)期分别为8±3h和7±3h(n=5);两者无显著性差异(p>0.05)。服药后24h的唾药浓度和血药浓度的比率为0.76±SD 0.17,回归方程:C_(血清)=0.9008C_(唾液)-0.2192,v=0.9765,p<0.001(n=30)。结果表明,用唾液浓度可替代血清浓度进行匹莫林的药物动力学研究。
The body concentration of five children with hyperkinetic syndrome after oral administration of Pamirin tablets was determined by high performance liquid chromatography (HPLC). About 2h after taking the drug, the plasma concentration and the concentration of saliva can reach the peak. The half-subtraction (decay) periods of blood and saliva were 8 ± 3h and 7 ± 3h respectively (n = 5). There was no significant difference between the two groups (p> 0.05). The ratio of saliva concentration to plasma concentration at 24 h after medication was 0.76 ± SD 0.17. Regression equation: C_ (serum) = 0.9008C_ (saliva) -0.2192, v = 0.9765, p <0.001 (n = 30). The results showed that pharmacokinetic studies of piroxicam were performed using saliva concentrations instead of serum concentrations.
