目的应用盐酸利多卡因致小鼠中毒惊厥模型,探讨硫酸镁对利多卡因中毒小鼠惊厥和死亡率的影响。方法 210只昆明小鼠,体重18~25g,按照随机数字表随机分为5组:生理盐水对照组、50mg/kg硫酸镁组、100mg/kg硫酸镁组、200mg/kg硫酸镁组、400mg/kg硫酸镁组,每组42只,分别预先腹腔注射4种剂量的硫酸镁或等容积的生理盐水,15min后腹腔注射100mg/kg利多卡因,观察小鼠惊厥潜伏期、持续期、惊厥发生率、死亡时间和死亡率。结果①与生理盐水组相比,硫酸镁可延长利多卡因致惊厥的潜伏期,缩短利多卡因致惊厥的持续时间,延长利多卡因致惊厥的死亡时间。②硫酸镁可以降低利多卡因致惊厥的发生率,200和400mg/kg硫酸镁组与生理盐水组比较,利多卡因致惊厥的发生率差异具有统计学意义(P﹤0.05);可降低利多卡因致惊厥的死亡率,50、100、200mg/kg硫酸镁组与生理盐水比较,利多卡因致惊厥的死亡率差异具有统计学意义(P﹤0.01,P﹤0.05)。③硫酸镁可降低利多卡因致惊厥的强度,随着硫酸镁剂量的增加,利多卡因致惊厥的等级逐渐降低。结论在本实验条件下,硫酸镁可拮抗利多卡因的致惊厥作用,降低利多卡因的毒性。
Objective To investigate the effect of magnesium sulfate on seizure and mortality in mice with lidocaine poisoning by using lidocaine hydrochloride-induced convulsion model in mice. Conclusion Under the experimental conditions, magnesium sulfate can antagonize the induced convulsion of lidocaine and reduce the toxicity of lidocaine.
