目的 研究DeltaPakC18反相填料用于P44纯化制备工艺可行性 ,为固相合成疏水性肽苗的制备工艺作探索性研究。方法 选用美国Waters公司DeltaPakC18反相色谱填料和高压液相色谱系统Delta6 0 0 ,采用乙腈、三氟乙酸体系为流动相 ,对P44从分析柱、半制备柱、径向加压制备柱 3个水平进行了基线分离载量及放大的研究。结果 目标肽峰面积积分分别为 71.14 %、70 .12 %、6 9.6 8% ,收率放大前后趋一致 ,载量分别为 10 0 μg、<5mg、<10mg ,制备目标肽收峰经SymmetryC18柱分析及PDA纯度鉴定为单一组分。结论 该填料可作为P44的小量精制 ,用作放大制备其载量尚不能满足需要
OBJECTIVE To study the feasibility of using DeltaPak C18 reversed-phase packing material for the purification of P44 and explore the preparation technology of hydrophobic peptide seedlings for solid-phase synthesis. Method DeltaPak C18 reversed-phase chromatography packing and high-pressure liquid chromatography system Delta6 0 0 from Waters Corporation of America were used. The mobile phase consisted of acetonitrile and trifluoroacetic acid was used as the mobile phase. P44 was separated from analytical column, semi-preparative column and radial pressurized preparative column Baseline separation of the load and amplification studies. Results The peak areas of the target peptides were 71.14%, 70.12% and 69.68%, respectively. The yields of the target peptides peaked at 10 0 μg, <5 mg and <10 mg, respectively. Analysis and PDA purity were identified as a single component. Conclusion The filler can be used as a small amount of refined P44, used to enlarge its capacity to prepare can not meet the needs
