目的:研究从温郁金中提取的β-榄香烯对人骨髓瘤RPMI-8226细胞增殖、周期和凋亡的影响。方法:采用MTT法检测细胞增殖;AnnexinV/PI双标流式术检测细胞周期、细胞凋亡;Hoechst33342/PI双染荧光显微镜观察β-榄香烯处理后骨髓瘤RPMI-8226细胞形态学变化。结果:β-榄香烯对RPMI-8226骨髓瘤细胞生长具有显著抑制作用,呈浓度和时间依赖性。与对照组相比G0/G1细胞比例显著升高,而S、G2/M细胞比例降低。β-榄香烯作用48h可诱导骨髓瘤细胞凋亡,凋亡率随药物浓度而递增。结论:β-榄香烯可有效抑制人骨髓瘤细胞增殖;其抑制作用与细胞周期阻滞和细胞凋亡激活有关。
OBJECTIVE: To study the effects of β-elemene extracted from warm turmeric on the proliferation, cell cycle and apoptosis of human myeloma RPMI-8226 cells. Methods: Cell proliferation was detected by MTT assay. Cell cycle and apoptosis were detected by Annexin V / PI double flow cytometry. Hoechst33342 / PI double staining was used to observe the morphological changes of myeloma RPMI-8226 cells after treatment with β-elemene. Results: β-elemene significantly inhibited the growth of RPMI-8226 myeloma cells in a concentration- and time-dependent manner. Compared with the control group, the proportion of G0 / G1 cells increased significantly, while the proportion of S, G2 / M cells decreased. β-Elemene 48h induced apoptosis of myeloma cells, apoptosis rate increased with the drug concentration. Conclusion: β-elemene can effectively inhibit the proliferation of human myeloma cells; its inhibitory effect is related to cell cycle arrest and activation of apoptosis.
