目的探讨亚急性砷暴露对小鼠海马各区星形胶质细胞的影响。方法将昆明种小鼠随机分为对照组及低、中和高剂量染毒组。以自由饮水方式饮含砷水,对照组小鼠饮蒸馏水。染毒8周后,心脏灌流固定,取大脑,进行胶质纤维酸性蛋白(GFAP)免疫组化染色,分析砷对小鼠海马各区GFAP表达情况,判断其对星形胶质细胞的影响。结果雌鼠海马CA1、CA3和DG区的GFAP积分光密度(IOD)值均先升高而后下降;在海马CA1和DG区,低剂量组的IOD值最高,并显著高于对照组,而高剂量组的IOD值则显著低于对照组;但在海马CA3区,低、中剂量组的IOD值均显著高于对照组,以中剂量组的IOD值最高,而高剂量组则显著低于对照组。雄鼠海马CA1和DG区GFAP的IOD值也先升高而后下降,中剂量组的IOD值最高,并显著高于对照组;但在海马CA3区,IOD值随染砷剂量的增加不断增高,高剂量组的IOD值最高,并显著高于对照组。结论低水平砷暴露可使小鼠海马星形胶质细胞活化,而高水平砷暴露会损伤海马星形胶质细胞;雌鼠海马星形胶质细胞对砷作用比雄鼠敏感;而无论雌、雄鼠,其海马CA1和DG区星形胶质细胞对砷的作用比CA3区敏感。
Objective To investigate the effects of sub-acute arsenic exposure on astrocytes in hippocampus of mice. Methods Kunming mice were randomly divided into control group and low, medium and high dose exposure groups. Arsenic drinking water with free drinking water, control mice drinking distilled water. After 8 weeks of exposure, the hearts were fixed by perfusion and the brain was taken for GFAP immunohistochemical staining. The expression of GFAP in the hippocampus of mice was analyzed to determine the effect of arsenic on the astrocytes. Results The GFAP integral optical density (IOD) values of hippocampal CA1, CA3 and DG increased first and then decreased in hippocampal CA1 and DG. The IOD values of hippocampal CA1 and DG were the highest and significantly higher than those in control group The IOD value of the dose group was significantly lower than that of the control group; but in the CA3 area of the hippocampus, the IOD values of the low and middle dose groups were significantly higher than those of the control group, the IOD value of the medium dose group was the highest, while the high dose group was significantly lower Control group. The IOD of GFAP in hippocampal CA1 and DG of male rats also increased first and then decreased, and the IOD of middle dose group was the highest, which was significantly higher than that of control group. However, the IOD value of hippocampal CA3 area increased with the increase of arsenic dose, The IOD value of the high-dose group was the highest, which was significantly higher than that of the control group. Conclusions Low level of arsenic exposure can activate hippocampal astrocytes in mice, while exposure to high levels of arsenic can damage hippocampal astrocytes. Hippocampal astrocytes in female rats are more sensitive to arsenic than male rats. However, , Male rats, the hippocampal CA1 and DG astrocytes on the role of arsenic than CA3 sensitive.
