目的观察海兔素(aplysin)对人乳腺癌细胞系MCF-7细胞增殖和血管内皮生长因子(VEGF)表达的抑制作用,以及对二甲基苯蒽(DMBA)诱导乳腺癌大鼠肿瘤组织中VEGF表达的抑制作用。方法用MTT法和ELISA法分别检测海兔素对MCF-7细胞增殖及VEGF表达的影响。用免疫组织化学法检测海兔素对DMBA诱导的大鼠乳腺癌组织中VEGF的影响。结果海兔素体外对MCF-7细胞有明显的增殖抑制作用,并伴随VEGF表达水平下降。体内可有效抑制大鼠乳腺癌组织中VEGF的表达,且与海兔素浓度有关。结论海兔素体外对MCF-7细胞具有明显的细胞毒作用并能抑制其VEGF的表达,体内可有效抑制乳腺癌组织中VEGF的表达。
Objective To observe the inhibitory effect of aplysin on the proliferation of human breast cancer cell line MCF-7 and on the expression of vascular endothelial growth factor (VEGF), and to investigate the effect of apysin on the tumor tissue of breast cancer induced by DMBA Inhibition of VEGF expression. Methods MTT and ELISA were used to detect the effect of laminin on the proliferation and the expression of VEGF in MCF-7 cells. Immunohistochemistry was used to detect the effect of sea-rabbit on VEGF in DMBA-induced rat breast cancer. Results The sea rabbit in vitro significantly inhibited the proliferation of MCF-7 cells, accompanied by decreased VEGF expression. The body can effectively inhibit the expression of VEGF in rat breast cancer tissue, and the concentration of sea rabbit. Conclusion The sea rabbit in vitro cytotoxic MCF-7 cells and can inhibit the expression of VEGF, in vivo can effectively inhibit the expression of VEGF in breast cancer tissue.
