建立苍耳子中原儿茶酸、新绿原酸、隐绿原酸、1,3-二咖啡酰奎宁酸的UPLC同时检测方法,测定和比较28批不同产地和集散地苍耳子中4种酚酸的含量。研究采用UPLC法的流动相为甲醇-0.1%磷酸水溶液,检测波长为220和327 nm,流速0.4 mL.min-1,柱温35℃。结果显示,原儿茶酸、新绿原酸、隐绿原酸和1,3-二咖啡酰奎宁酸分别在0.357 0~35.70、2.500~250.0、1.060~106.1和1.010~101.0μg.mL-1内具有良好的线性关系(r2=0.999 9),平均回收率分别为(n=6)97.68%、99.55%、97.92%和100.4%。该方法快速、准确、重复性好,可为苍耳子的质量控制提供快速准确的检测方法。
To establish a simultaneous UPLC method for the determination of Xanthium sibiricum, neogenic chlorogenic acid, chlorogenic acid and 1,3-dicaffeoylquinic acid in Xanthium sibiricum, and to determine and compare the contents of four kinds of phenols in 28 batches of Xanthium sibiricus Acid content. The mobile phase using UPLC was methanol-0.1% phosphoric acid solution. The detection wavelength was 220 and 327 nm, the flow rate was 0.4 mL.min-1 and the column temperature was 35 ℃. The results showed that protocatechuic acid, neogenuric acid, cryptocholic acid and 1,3-dicaffeoylquinic acid were in the range of 0.357 0 to 35.70, 2.500 to 250.0, 1.060 to 106.1 and 1.010 to 101.0 μg.mL-1 (R2 = 0.999 9). The average recoveries were 97.68%, 99.55%, 97.92% and 100.4% respectively (n = 6). The method is rapid, accurate and reproducible, and can provide a fast and accurate method for the quality control of Xanthium ssp.
