目的:通过研究去卵巢骨质疏松症大鼠股骨中GPR48和CREB的蛋白表达,探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究左归丸的作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制骨质疏松症模型,采用左归丸对实验大鼠治疗12周,以盖天力片作为阳性对照组,正常组、假手术组作为标准对照组,模型组作为空白对照,用ELISA法检测骨质疏松症大鼠股骨GPR48和CREB的蛋白表达。结果:ELISA方法检测表明:正常大鼠股骨中存在GPR48和CREB的蛋白表达。模型组大鼠股骨中GPR48和CREB的蛋白表达水平有明显的下降。左归丸组、盖天力组用药12周后,与模型组相比较,左归丸组可显著上调股骨中GPR48和CREB的蛋白表达水平。结论:1正常大鼠股骨中存在GPR48和CREB的蛋白表达;2股骨中GPR48和CREB的蛋白表达下降可能是导致骨质疏松症发生的机理之一;3左归丸能够上调GPR48和CREB的蛋白表达,表明左归丸对骨质疏松症有一定的防治作用。
OBJECTIVE: To investigate the protein expression of GPR48 and CREB in the femur of ovariectomized rats with osteoporosis, and explore the pathological mechanism of osteoporosis induced by kidney removal in ovariectomized rats and the mechanism of Zuogui Pill. Methods: Osteoporosis model was induced by bilateral ovary removal in female rats. The rats in Zuogui Pill group were treated with Zuogui Pill for 12 weeks. Gatip was used as positive control group, normal group and sham operation group as standard control group , The model group as a blank control, the protein expression of GPR48 and CREB in the femur of osteoporosis rats was detected by ELISA. Results: The results of ELISA showed that the protein expression of GPR48 and CREB existed in the normal femur. The protein expression of GPR48 and CREB in the femur of the model group decreased significantly. Compared with model group, Zuoguiwan group could significantly upregulate the protein expression of GPR48 and CREB in Zuoguiwan group and GTI group. CONCLUSION: 1 There is expression of GPR48 and CREB protein in normal femur. The decrease of protein expression of GPR48 and CREB in 2 femur may be one of the mechanisms leading to osteoporosis.3. Zuogui pill can upregulate the protein of GPR48 and CREB Expression, indicating that Zuogui Wan on osteoporosis have a preventive effect.
