目的 研究大鼠局灶性脑缺血不同缺血时间皮质半暗带和中心区葡萄糖转运子 3(GL UT3 )转录水平的表达规律。方法 用插线法建立大鼠局灶性脑缺血模型 ,剥取缺血半暗带及中心区皮质组织 ,采用逆转录 -聚合酶链反应 (RT- PCR) ,测定 GL U T3 m RNA水平的变化。结果 缺血半暗带 GL U T3 m RNA在缺血 3小时升高 ,2 4小时达到高峰 ,96小时后基本恢复正常。缺血中心区 GL U T3 m RNA在缺血后 3小时有一短暂的升高 ,随后迅速下降呈低水平表达。结论 GL UT3 在缺血半暗带的表达上调 ,有可能是机体对缺血损伤的保护性反应。
Objective To investigate the transcriptional regulation of GLUT3 in the cortex penumbra and central area of rats with focal cerebral ischemia and ischemia at different time points. Methods The model of focal cerebral ischemia in rats was established by patch-line method. The ischemic penumbra and central cortex tissues were removed and the level of GL U T3 m RNA was determined by reverse transcription-polymerase chain reaction (RT-PCR) The change. Results The ischemic penumbra GL U T3 m RNA increased at 3 hours after ischemia, peaked at 24 hours and returned to normal after 96 hours. GL U T3 m RNA in the ischemic center had a transient increase 3 hours after ischemia and then rapidly decreased to a low level. CONCLUSION: GLUT3 is up-regulated in ischemic penumbra and may be a protective response to ischemic injury.
