建立了八角中莽草酸的毛细管区带电泳—紫外检测分析新方法.在单一磷酸盐和硼砂溶液中由于电扩散现象,莽草酸峰宽并严重前倾或脱尾,采用磷酸盐—硼砂混合溶液为缓冲液改善了峰形并提高了检测灵敏度,通过优化获得其最佳分离条件为pH 8.63,10 mmol/L NaH_2PO_4-9 mmol/L硼砂,分离电压25 kV.该条件下莽草酸的峰面积与浓度呈良好的线性关系,线性方程的相关系数为0.9998,检测限(S/N=3)为0.12μg/mL,比文献报道的方法约高10倍.该方法成功用于八角壳和籽中莽草酸的定量分析,其加标回收率分别为98.9%和104.6%.
A new method of capillary zone electrophoresis-ultraviolet detection analysis has been established for octanoic oxalic acid. In the single phosphate and borax solution, shikimic acid has a broad peak and leans forward or tails heavily because of the electric diffusion phenomenon. Phosphate-borax mixed solution is used. The peak shape was improved for the buffer and the detection sensitivity was improved. The optimal separation conditions were optimized to be pH 8.63, 10 mmol/L NaH2PO4-9 mmol/L borax, and the separation voltage was 25 kV. The peak area of shikimic acid under this condition was obtained. There was a good linear relationship with the concentration. The correlation coefficient of the linear equation was 0.9998, and the detection limit (S/N=3) was 0.12 μg/mL, about 10 times higher than the method reported in the literature. This method was successfully applied to octagonal shells and seeds. Quantitative analysis of scoxic acid, the spike recovery rate were 98.9% and 104.6%, respectively.
