观察偏心旋转刺激下前庭核早期即刻基因c fos的表达特征和抗运动病药物对这种表达的影响。方法取SD大鼠 1 9只分为对照组、旋转组、药物组三组。旋转组用偏心旋转刺激方式刺激60min ;药物组在偏心旋转刺激前 45min腹腔注射抗运动病药物盐酸扁桃桥哌酯 (PAPM ) ;对照组不进行旋转刺激。用免疫组化法和图象分析技术对大鼠脑干前庭神经fos蛋白含量变化进行定量、定位测定。结果偏心旋转刺激后前庭核团细胞核中fos蛋白含量明显增加 ,而PAPM并不影响fos蛋白的表达。结论前庭核的c fos表达是前庭神经系统对外界刺激应答的一种方式 ;且这种表达不直接参与运动病的发生和发展。
To observe the expression characteristics of c fos gene in early vestibular nucleus and the effects of anti-motion sickness drugs on this expression under eccentric rotation stimulation. Methods Nineteen SD rats were divided into control group, rotation group and drug group. Rotation group was stimulated by eccentric rotary stimulation for 60min. In the drug group, antimyocardial drug Pilaster Histamine Hydrochloride (PAPM) was injected intraperitoneally 45min before eccentric rotation stimulation; the control group was not subjected to rotational stimulation. Immunohistochemistry and image analysis techniques were used to quantify and locate fos protein in rat brainstem vestibular nucleus. Results The fos protein content in nucleus of vestibular nuclei increased obviously after eccentric rotation stimulation, while PAPM did not affect the expression of fos protein. Conclusion The c fos expression in the vestibular nucleus is a way of the vestibular nervous system responding to external stimuli. And this expression is not directly involved in the occurrence and development of motion sickness.
