目的:观察内毒素对肝细胞的影响及丹参素的保护作用。方法:采用离体肝细胞原代培养技术,观察不同浓度内毒素及加入丹参素对肝细胞几项指标的影响。结果:内毒素可使乳酸脱氢酶(LDH)漏出增多,同时培养液上清中超氧化物歧化酶(SOD)减少,一氧化氮(NO)增加。应用丹参素后,可部分逆转这些改变。结论:内毒素可直接导致肝细胞损伤,丹参素可通过增强SOD活力,减少NO的产生而发挥其保护作用。
Objective: To observe the effect of endotoxin on hepatocytes and the protective effect of Danshensu. Methods: The primary culture technique of isolated hepatocytes was used to observe the effects of different concentrations of endotoxin and danshensu on several indexes of hepatocytes. RESULTS: Endotoxins increased leakage of lactate dehydrogenase (LDH), and superoxide dismutase (SOD) in culture supernatants decreased, and nitric oxide (NO) increased. After application of Danshensu, these changes can be partially reversed. Conclusion: Endotoxin can directly lead to hepatocyte injury. Danshensu can exert its protective effect by increasing SOD activity and decreasing NO production.
