目的研究石榴健胃片对脾肾阳虚型溃疡性结肠炎(SKD-UC)模型大鼠结肠肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6和IL-10的影响。方法采用15%大黄混悬液灌胃联合2,4,6-三硝基苯磺酸-乙醇溶液灌肠的病证结合法建立SKD-UC大鼠模型,将模型大鼠随机分为阳性组,模型组,石榴健胃片高、中、低剂量组,每组20只,另取20只作为空白组。造模成功后第2天开始进行药物干预,阳性组大鼠灌胃柳氮磺胺吡啶(SASP,0.36g/kg),石榴健胃片高、中、低剂量组分别灌胃石榴健胃片混悬液2.4,1.2,0.6g/kg,模型组和空白组灌胃等体积生理盐水,每天1次,连续21d。末次给药后24h处死大鼠,取结肠组织,采用逆转录-聚合酶链反应(RT-PCR)法检测结肠组织中IL-1β,IL-6,IL-10,TNF-α的基因表达。结果与空白组比较,模型组大鼠结肠IL-1β,IL-6,TNF-α的mRNA相对表达量明显增高,IL-10的mRNA相对表达量明显降低,组间比较差异均有高度统计意义(P<0.01);与模型组比较,石榴健胃片高、中、低剂量组大鼠结肠IL-1βmRNA和TNF-αmRNA相对表达量明显降低(P<0.05或P<0.01),高、中剂量组IL-6mRNA相对表达量明显降低(P<0.01),而高、中、低剂量组IL-10mRNA相对表达量明显升高(P<0.05或P<0.01);与阳性组比较,石榴健胃片高剂量组大鼠结肠IL-1β,TNF-α和IL-6的mRNA相对表达量及中剂量组IL-1βmRNA相对表达量明显降低(P<0.05),高、中剂量组IL-10mRNA相对表达量明显升高(P<0.05);各剂量组之间IL-1β,IL-6,TNF-α和IL-10的mRNA相对表达量比较差异均无统计意义(P>0.05)。结论石榴健胃片对SKD-UC模型大鼠的结肠TNF-α,IL-1β,IL-6及IL-10的基因表达具有调节作用。
Objective To study the effects of Pomegranate Tablets on the expressions of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), IL-6 and IL-8 in colon and kidney of rats with spleen and kidney yang deficiency type ulcerative colitis -10 of the impact. Methods The model of SKD-UC rats was established by the combination of the disease-syndrome method of intragastric administration of 15% rhubarb suspension and enema of 2,4,6-trinitrobenzene sulfonic acid-ethanol solution. The model rats were randomly divided into positive group, Model group, pomegranate stomach tablet high, medium and low dose group, each group of 20, another 20 as a blank group. Medication was started on the second day after successful model establishment. Rats in the positive group were treated with SASP (0.36g / kg, SASP, 0.36g / kg). Pomegranate stomach tablet high, Suspension 2.4,1.2,0.6 g / kg, the model group and the blank group intragastric administration of equal volume of normal saline once a day for 21 days. The rats were sacrificed 24h after the last administration, and the colon tissues were harvested for detection of the gene expression of IL-1β, IL-6, IL-10 and TNF-α by RT-PCR. Results Compared with the blank group, the mRNA expression of IL-1β, IL-6 and TNF-α in the model group was significantly increased, while the relative expression of IL-10 mRNA was significantly decreased in the model group (P <0.01) .Compared with the model group, the relative expression of IL-1βmRNA and TNF-αmRNA in the colon of high, medium and low doses of Pomegranate Tablets decreased significantly (P <0.05 or P <0.01) Compared with the positive group, the relative expression of IL-6mRNA was significantly decreased (P <0.01), while the relative expression of IL-10mRNA in high, middle and low dose groups was significantly increased (P <0.05 or P <0.01) The relative mRNA expression of IL-1β, TNF-α and IL-6 in colonic tissue of high-dose stomach group and the relative expression of IL-1βmRNA in medium dose group were significantly decreased (P <0.05) The relative expression of IL-1β, IL-6, TNF-α and IL-10 in each dose group was significantly higher than that in other groups (P <0.05). Conclusion Pomegranate Stomach Tablet can modulate the gene expression of TNF-α, IL-1β, IL-6 and IL-10 in colon of SKD-UC rats.
