目的研究胰岛素样生长因子Ⅰ型受体(IGF-IR)激酶抑制剂NVP-AEW-541及阿霉素联用对肝癌细胞株smmc-7721的治疗效果。方法 NVP-AEW-541与阿霉素单独和联合应用肝癌细胞株smmc-7721后,MTT法检测细胞生长抑制率,流式细胞术检测细胞凋亡,Western blot检测p27、抗凋亡蛋白Bcl-2及促凋亡蛋白Bax的表达。结果 NVP-AEW-541和阿霉素均可诱导smmc-7721细胞凋亡。与单独使用相比,两药联用对smmc-7721细胞抑制作用有明显提高(P<0.05),细胞凋亡率增加(P<0.05),对增加Bax、p27的表达及抑制Bcl-2的表达更明显。结论 NVP-AEW-541和阿霉素均可抑制smmc-7721细胞的增殖、诱导细胞凋亡,在一定浓度范围内,呈量-效关系。
Objective To investigate the therapeutic effect of insulin-like growth factor type 1 receptor (IGF-IR) kinase inhibitor NVP-AEW-541 and doxorubicin on human hepatocellular carcinoma cell line smmc-7721. Methods The inhibitory rate of cell growth was detected by MTT assay. The apoptosis rate of p27 and Bcl-541 were detected by flow cytometry. The expression of NVP-AEW-541 and doxorubicin alone and in combination with smmc- 2 and pro-apoptotic protein Bax. Results Both NVP-AEW-541 and doxorubicin induced the apoptosis of smmc-7721 cells. The inhibitory effect of the combination of the two drugs on smmc-7721 cells was significantly increased (P <0.05) and the apoptosis rate was increased (P <0.05), compared with the single use, the expression of Bax and p27 and the inhibition of Bcl-2 The expression is more obvious. Conclusion Both NVP-AEW-541 and doxorubicin can inhibit the proliferation and induce the apoptosis of smmc-7721 cells in a dose-dependent manner within a certain concentration range.
