目的研究子宫颈癌Caski细胞株中,胰岛素样生长因子-2(IGF-2)刺激细胞增殖的同时,对钾氯离子协同转运子(kcc)基因表达的调节作用。方法2006年1月至8月于中国医科大学盛京医院院中心实验室,采用酶联免疫吸附实验法确定不同时间点IGF-2细胞增殖曲线,采用聚合酶链反应法比较不同浓度、不同时间IGF-2作用下kcc1、kcc3、kcc4 mRNA表达水平的差异。结果低浓度的IGF-2刺激Caski细胞增殖的同时抑制细胞表面kcc基因的表达,kcc1、kcc3、kcc4 mRNA分别下降30%、50%和80%,随着IGF-2浓度的升高,对kcc基因的作用逐渐减弱。结论IGF-2对Caski细胞表面的kcc基因表达存在影响,这种刺激作用呈时间和浓度依赖性。为进一步研究IGF2通过kcc参与子宫颈细胞恶变过程提供了实验依据。
Objective To study the regulatory effect of insulin-like growth factor-2 (IGF-2) on cell proliferation in cervical cancer Caski cell line and on the expression of potassium-chloride synergistic transporter (kcc) gene. Methods From January to August 2006, the proliferation of IGF-2 cells at different time points was determined by enzyme-linked immunosorbent assay (ELISA) in the laboratory of Shengjing Hospital of China Medical University from January to August 2006. The effects of different concentrations and different time The difference of kcc1, kcc3 and kcc4 mRNA expression by IGF-2. Results Low concentration of IGF-2 stimulated the proliferation of Caski cells and inhibited the expression of kcc gene on the cell surface. The expression of kcc1, kcc3 and kcc4 mRNA decreased by 30%, 50% and 80% respectively. With the increase of IGF-2, The role of genes gradually weakened. Conclusion IGF-2 has an effect on the expression of kcc gene on the surface of Caski cells. The stimulation is time-and concentration-dependent. To further study the IGF2 by kcc involved in cervical malignant process provides an experimental basis.
