目的研究血管生成抑制剂TNP-470对人肝癌高转移细胞株MHCC97-H体外形成血管生成拟态的影响。方法TNP-470作用于MHCC97-H细胞,应用MTT试验、体外侵袭实验,检测MHCC97-H细胞增殖活性和侵袭力的变化。建立MHCC97-H细胞人工基底膜基质凝胶体外三维细胞培养模型,观察MHCC97-H细胞能否形成血管生成拟态以及TNP-470对其影响,应用扫描电镜、透射电镜观察细胞结构改变。结果TNP-470对MHCC97-H细胞增殖无显著抑制作用,可抑制其体外侵袭能力,明显抑制MHCC97-H细胞形成血管生成拟态,减少细胞表面的微绒毛和细胞间连接。结论人肝癌高转移细胞株MHCC97-H可形成血管生成拟态,TNP-470能抑制人肝癌细胞株MHCC97-H体外血管生成拟态形成。
Objective To study the effect of angiogenesis inhibitor TNP-470 on the formation of angiogenic mimicry in human hepatocellular carcinoma cell line MHCC97-H in vitro. Methods TNP-470 was used to act on MHCC97-H cells. MTT assay and in vitro invasion assay were performed to detect the changes of proliferation and invasiveness of MHCC97-H cells. The establishment of MHCC97-H cells in vitro three-dimensional cell culture matrix gel matrix gel culture model to observe the MHCC97-H cells can form vasculogenic mimicry and TNP-470 on its impact, the application of scanning electron microscopy, transmission electron microscopy changes in cell structure. Results TNP-470 had no significant inhibitory effect on the proliferation of MHCC97-H cells, which could inhibit the invasion ability of MHCC97-H cells in vitro and significantly inhibit the formation of angiogenic mimicry and decrease the microvilli and intercellular junctions on MHCC97-H cells. Conclusion Human hepatocellular carcinoma cell line MHCC97-H can form mimicry of angiogenesis. TNP-470 can inhibit the mimicry of human hepatocellular carcinoma cell line MHCC97-H in vitro.
