目的研究甘肃栽培甘草内生菌有效菌株发酵液与宿主水煎液、总黄酮提取物、总皂苷提取物对脂多糖(LPS)刺激的raw264.7细胞炎症模型分泌炎症因子的调节作用。方法采用Griess法、双抗夹心法,检测甘肃栽培甘草内生菌有效菌株发酵液与宿主水煎液、总黄酮提取物、总皂苷提取物对脂多糖(LPS)刺激raw264.7细胞分泌NO、TNF-α、IL-6的含量的变化。结果经LPS刺激后,与模型组相比,栽培甘草的水煎液和栽培甘草的总黄酮、总皂苷以及其有效菌株的发酵液均能抑制NO、TNF-α、IL-6的分泌(P<0.05)。结论甘草内生菌JTZB006、JTZB014、JTZB016、JTZB018、JTZB062的发酵液有抑制raw264.7分泌NO、TNF-α、IL-6的作用。
Objective To study the regulation of inflammatory factors secreted by lipopolysaccharide (LPS) -stimulated raw264.7 cells in the culture of Glycyrrhiza uralensis Fisch. Methods Griess method and double-antibody sandwich method were used to detect the effect of LPS on the secretion of nitric oxide (NO) in raw264.7 cells stimulated by Lactobacillus plantarum, TNF-α, IL-6 content changes. Results After stimulated by LPS, compared with the model group, the decoction of licorice and the total flavonoids and total saponins of cultivated licorice and the fermentation broths of their effective strains all inhibited the secretion of NO, TNF-α and IL-6 (P <0.05). Conclusion The fermentation broth of endotoxin of JTZB006, JTZB014, JTZB016, JTZB018 and JTZB062 could inhibit the secretion of NO, TNF-α and IL-6 by raw264.7.
