目的探究脑缺血模型大鼠经30d高压氧治疗对大脑神经细胞的影响。方法成年SD大鼠随机分为三组:正常对照组、缺血再灌注(IR)组、高压氧治疗(HBO)组。IR组和HBO组动物经大脑中动脉栓塞(MCAO)1.5h后再灌注。所有大鼠均于术后2h腹腔注射BrdU(50mg/kg体重),连续2次,间隔24h。HBO组高压氧治疗(2.8个绝对大气压,60min)30d,每日一次。所有大鼠于31d麻醉处死,取脑,切片,进行Nestin和BrdU的免疫组织化学染色,光镜观察并于损伤区取图。结果 HBO组和IR组Nestin+和BrdU+细胞表达于损伤区,两者密度在HBO组中均显著高于IR组(P<0.05),而正常对照组均未见阳性结果。结论 30dHBO治疗可促进大鼠缺血脑组织神经细胞增殖或对其有保护作用。
Objective To investigate the effects of hyperbaric oxygen on cerebral neurons in rats with cerebral ischemia after 30 days. Methods Adult SD rats were randomly divided into three groups: normal control group, ischemia reperfusion (IR) group and hyperbaric oxygen therapy (HBO) group. Animals in IR group and HBO group were reperfused 1.5 hours after middle cerebral artery occlusion (MCAO). All rats were intraperitoneal injection of BrdU (50mg / kg body weight) 2h after operation, 2 times in a row, separated by 24h. HBO group hyperbaric oxygen therapy (2.8 absolute atmospheric pressure, 60min) 30d, once daily. All rats were anesthetized and sacrificed on day 31. The brain and sections were taken for immunohistochemical staining of Nestin and BrdU. The specimens were observed under light microscope and taken in the injured area. Results Nestin + and BrdU + cells in HBO group and IR group were expressed in the injured area, both of which were significantly higher in HBO group than in IR group (P <0.05). No positive results were found in normal control group. Conclusion 30 dHBO treatment can promote the proliferation of neuronal cells in ischemic brain tissue or its protective effect in rats.
