为探讨采后贮期莲雾果实中腺苷酸含量以及能荷变化对果肉组织絮状绵软的影响,建立高效液相色谱法测定采后台湾“黑珍珠”莲雾果实中ATP、ADP和AMP含量的方法。采用Atlantis T3柱(5.0μm,4.6 mm×250mm,Waters),流动相为磷酸盐缓冲液(20 mmol/L KH2PO4:20 mmol/L Na2HPO4,pH 7.0),流速1.2 mL/min,柱温30℃,检测波长254 nm条件,结果 ATP、ADP和AMP在6.25~200μg/mL质量浓度范围呈现良好的线性关系。用沸水提取的样品中3种腺苷酸ATP、ADP、AMP的稳定性较好,加标回收率分别为93.71%(RSD=1.74%)、97.28%(RSD=3.42%)、98.17%(RSD=3.12%)。该方法具有较高的灵敏度与精密度,重现性好,可用于测定采后莲雾果实中的腺苷酸含量。
