目的优化肺鳞癌组织双向电泳方法,进行蛋白图谱差异分析。方法收集癌组织以及癌外周组织标本,通过优化参数和方法,应用双向电泳技术分离组织总蛋白,比较差异蛋白。用基质辅助激光解吸电离飞行时间质谱得到相应的肽质指纹图谱(PMF),并进一步检索数据库。结果获得了分辨率高、重复性好的双向电泳图谱,得到了若干与细胞增生、分化、周期调控或肿瘤发生有关的蛋白。结论建立了肺鳞状细胞癌组织的双向电泳方法。
Objective To optimize two-dimensional gel electrophoresis of squamous cell carcinoma of the lung and analyze the difference of protein profiles. Methods Tissue samples of cancerous tissues and peripheral tissues of cancer were collected. Two-dimensional gel electrophoresis was used to separate the total proteins in the tissues and the differentially expressed proteins by optimizing parameters and methods. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry was used to obtain the corresponding peptide fingerprint (PMF) and to further search the database. The results obtained two-dimensional gel electrophoresis with high resolution and good repeatability, and obtained a number of proteins related to cell proliferation, differentiation, cycle regulation or tumorigenesis. Conclusion A two-dimensional electrophoresis method was established for lung squamous cell carcinoma.
