目的评价CO2气腹及不同气腹压力对裸鼠腹腔种植肿瘤基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达的影响。方法 30只裸鼠建立腹腔种植肿瘤模型,造模成功后随机分组:对照组、5mmHg和10mmHgCO2气腹组,气腹作用时间1h。气腹干预后1周检测肿瘤组织MMP2、TIMP-1蛋白表达情况。结果与对照组比较,气腹干预后1周5mmHg和10mmHgCO2气腹组肿瘤组织MMP-2蛋白表达均显著升高(P<0.01),且10mmHg组高于5mmHg组(P<0.01)。两组TIMP-1表达降低,且10mmHg组低于5mmHg组(P<0.01)。MMP-2/TIMP-1比例下降,10mmHg组下降更为显著(P<0.01)。结论 :CO2气腹作用后早期可使肿瘤组织MMP-2升高,TIMP-1下降,气腹压力高作用更为明显。
Objective To evaluate the effect of CO2 pneumoperitoneum and different pneumoperitoneum pressure on the expression of tumor necrosis factor-α (MMP-2) and matrix metalloproteinase-1 (TIMP-1) in peritoneal cavity of nude mice. Methods Twenty nude mice were randomly divided into control group, pneumoperitoneum group of 5mmHg and 10mmHgCO2, and pneumoperitoneum for 1h. One week after pneumoperitoneum intervention, the expression of MMP2 and TIMP-1 protein in tumor tissue was detected. Results Compared with the control group, the expression of MMP-2 protein in the pneumoperitoneum group at 5 mmHg and 10 mmHgCO2 pneumoperitoneum was significantly increased after pneumoperitoneum intervention (P <0.01), and higher in the 10 mmHg group than in the 5mmHg group (P <0.01). The expression of TIMP-1 decreased in both groups, and the level in 10mmHg group was lower than that in 5mmHg group (P <0.01). MMP-2 / TIMP-1 ratio decreased, 10mmHg group decreased more significantly (P <0.01). CONCLUSION: Early CO2 pneumoperitoneum can increase MMP-2, decrease TIMP-1 and increase pneumoperitoneum pressure.
