心脏组织石蜡大切片法我室在克山病研究工作中为进行病灶立体重建,在一张切片上全面地观察整个心脏的病灶分布,我们试作了大块心脏及整个小儿心脏石蜡切片。由于克山病心肌癜痕组织较多,经酒精脱水,二甲苯透明后其硬度更强,加以组织块大,给切片带来了较大的困难,为克服这些难点,我们在软化组织、充分透阴、改进??木块等方面采取了措施,而成功地在Rechard滑坳式切片机上切出7×5×O.5公分的小儿整个心脏,厚度为5—8微米,染色种类为HE,VG结缔组织、Mallory氏三色胶元纤维以及磷钨酸苏木素横纹肌。兹将所用的脱水包埋流程分述如下:冲水12小时,5%石炭酸软化24小时,水洗4小时,85%酒精15小时,95%酒精I 9小时,95%酒精I 15小时,100%洒精I 4小时,100%酒精I 4小时,酒精二甲苯I液1小时,二甲苯I 1小时,石蜡I4~6小时,石蜡I 4~6小时。为弥补脱水前软化不足,可以在切片前用70%酒精甘油-石炭酸直接浸泡蜡块2~7天。此外,为保证充分脱水,故采用了较长时间的酒精脱水。在室温下二次二甲苯透明,必须以肉眼控制其透明程度。在切片时进刀必须缓慢,用力均匀,可以采用锐角进刀以减少阻力。
