本文采用培养心肌细胞药物性心律紊乱模型,观察描记到炙甘草注射液具有明显的负性频率作用而无正性肌力作用;在加大剂量时细胞簇的搏动频率和幅度随药物剂量的增加而下降,当药物剂量增加到有效浓度3倍以上时有90%的搏动点出现停搏现象。对哇巴因、乌头碱、地高辛、氯化钙四种药物均因K~+、N~+、CA~+离子浓度发生变化而造成的心肌细胞节律紊乱,炙甘草注射液有明显的对抗作用。
In this study, the model of drug-induced heart rhythm disturbance in cultured cardiomyocytes was used to observe that Zhigancao injection had obvious negative frequency effect without positive inotropic effect; the frequency and amplitude of beating of cell clusters increased with the increase of drug dose when the dose was increased. However, when the drug dose increased to more than 3 times the effective concentration, 90% of the beat points appeared to be arrested. For ouabain, aconitine, digoxin, and calcium chloride, the cardiomyocyte rhythm disorder caused by changes in the concentration of K~+, N~+, and CA~+ ions was significantly increased.
